A Urinary Proteomics Approach to Predict Treatment Response in Acute Antibody-Mediated Rejection

نویسندگان

چکیده

Acute antibody-mediated rejection (AMR) is a severe complication affecting kidney allografts' long-term survival. Timely detection and appropriate treatment of AMR are crucial for improving graft outcomes. This study aimed to discover non-invasive urinary biomarkers that can predict the response therapy in patients with AMR. Materials Methods: In this case-control study, urine samples from 21 biopsy-proven underwent proteomics using label-free quantification. The were divided into two groups: responders non-responders based on their function. Urinary proteins identified, expressions compared between groups identify potential candidate biomarkers. Out 1020 identified proteins, 257 found be differentially expressed groups. Among these, 153 showed increased expression non-responder patients, while 104 decreased expression. Non-responder exhibited higher activation complement pathway extracellular matrix degradation than responders. Insulin-like growth factor binding protein 6 (IGFBP-6) emerged as most sensitive specific biomarker predicting non-response Our has successfully proteome distinguish These associated various biological processes reflect pathogenesis severity Further research necessary validate these findings.

Acute antibody-mediated rejection after intestinal transplantation

AIM To investigate the incidence, risk factors and clinical outcomes of acute antibody-mediated rejection (ABMR) after intestinal transplantation (ITx). METHODS A retrospective single-center analysis was performed to identify cases of acute ABMR after ITx, based on the presence of donor-specific antibody (DSA), acute tissue damage, C4d deposition, and allograft dysfunction. RESULTS Acute AB...

Acute antibody-mediated rejection after lung transplantation.

BACKGROUND Antibody-mediated rejection (AMR) after lung transplantation remains enigmatic, and there is no consensus on the characteristic clinical, immunologic and histologic features. METHODS We performed a retrospective, single-center cohort study and identified cases of acute AMR based on the presence of circulating donor-specific human leukocyte antigen (HLA) antibodies (DSA), histologic...

Antibody-mediated rejection.

The introduction of both complement 4d (C4d) staining in renal allograft biopsies and sensitive methods to detect anti-human leukocyte antigen antibodies, such as single antigen bead flow assays, into tissue-typing techniques have shown the importance of antibody-mediated alloimmune response in kidney transplantation. The use of these sensitive methods, combined with the increased number of tra...

Bortezomib-based treatment of acute antibody-mediated rejection: a case report.

Antibody-mediated rejection (AMR) is an important factor affecting survival after renal transplantation. A highly selective proteasome inhibitor, bortezomib, clears activated plasma cells from the body and has important therapeutic effect on AMR. We investigated the effects of bortezomib on AMR in a patient after a second renal transplant. Biopsy confirmed the diagnosis of mixed cellular reject...
